Start by matching the requested measurement to the test: clotting-time testing differs from fibrinogen measurement, and both differ from Factor V Leiden, Protein C, or Protein S testing. Clotting times and fibrinogen answer different questions
PT and INR measure a clotting-time pathway and are commonly considered together; INR standardizes the PT result for comparison. Activated PTT measures a different clotting-time pathway, so the Prothrombin Time (PT) (INR) and Partial Thromboplastin Time (PTT) test reports all three values. Fibrinogen Activity measures functional fibrinogen using the Clauss method. Fibrinogen is a protein used in clot formation, so its activity result addresses a different part of the process than PT, INR, or activated PTT. A result outside the reference interval does not identify one cause by itself and should be interpreted with the reason testing was ordered.
D-dimer, inherited factors, and natural anticoagulants
D-Dimer with CBC reports quantitative D-dimer alongside red blood cell, white blood cell, hemoglobin, hematocrit, and platelet measurements. D-dimer reflects fibrin breakdown and is not specific to one condition, which is why symptoms and clinical history matter. Factor V (Leiden) Mutation Analysis looks specifically for the named mutation rather than measuring clotting time. Protein C Activity measures the activity of Protein C, while Protein S Antigen, Free measures the free Protein S antigen level. These are distinct measurements used in clotting-risk evaluations; one result alone does not establish the reason for a clotting concern.
